Nanoparticle delivery of siRNA against TWIST to reduce drug resistance and tumor growth in ovarian cancer models

siRNA targeting TWIST can be complexed by electrostatic interactions with either a third generation polyamidoamine (PAMAM) dendrimer micelle or polyethyleneimine (PEI) coated mesopourous silica nanoparticle (MSN). Delivery of the siRNA payload into Ovcar8 cells resulted in substantial TWIST knockdown and sensitization of cells to cisplatin treatment. In vivo studies demonstrated that MSNs carrying TWIST siRNA combined with cisplatin produced greater reduction in tumor burden than cisplatin alone.123
