Structural revision of periplocosides and periperoxides, natural immunosuppressive agents from the genus Periploca

► The structural revision of pregnane glycosides are described, including use of X-ray crystallographic diffraction analysis. ► The peroxy function in these pregnane glycosides were revised to orthoester groups. ► The orthoester group is critical for the immunosuppressive activity.
