Plasmid DNA recovery from fermentation broths by a combined process of micro- and ultrafiltration: Modeling and application

▸ Plasmid DNA (pDNA) recovery from fermentation broths was investigated. ▸ High plasmid recovery yields were obtained by microfiltration, after cell lysis. ▸ Small fragments of genomic DNA (gDNA) re-dissolved from the suspended solids. ▸ pDNA was purified from RNA, proteins and the gDNA fragments by ultrafiltration. ▸ Permeation of pDNA, RNA and gDNA fragments was modeled on a theoretical basis.
