A monolith purification process for virus-like particles from yeast homogenate

► Separation of HBsAg virus-like particles from yeast is achieved with a monolith. ► Dynamic binding capacity for VLP with untreated feed is 3-4x that of a packed bed. ► Confocal microscopy is used to visualise the degree and position of lipid fouling. ► A lipid layer is seen on the monolith even after regeneration with 30% isopropanol. ► Reducing the lipid in the yeast homogenate causes the capacity to increase 2.5x.
