Discovery of potent N-(isoxazol-5-yl)amides as HSP90 inhibitors

By fragment screening and x-ray crystallography techniques a scaffold of 4,5-diarylisoxazole was selected and developed to a novel series of HSP90 inhibitors with excellent in vivo activities.
