► The membrane topology of the HCV NS4B protein is described. ► HCV NS4B has five transmembrane domains and possibly a sixth one. ► The sixth possible transmembrane domain of NS4B could behave as a viral pre-transmembrane segment. ► These segments should be involved in the formation of the replication complex.
