► Characterization of steam-exploded spent Shiitake mushroom medium was evaluated. ► Spent Shiitake mushroom medium can be used as a substrate of alcohol fermentation. ► Water extraction after steam explosion is desired for alcohol fermentation of biomass. ► Steam explosion at 25 atm and 5 min is optimal for alcohol fermentation of biomass.
