► MSL cis-spreading from the roX gene upregulates the neighboring genes. ► The roX-neighboring genes are upregulated by approximately 2-fold. ► Haplo-insufficient phenotypes of the Minute mutant are rescued by MSL cis-spreading. ► Upregulation by MSL cis-spreading is a general mechanism in several tissue types.
