Expression, purification, and characterization of coiled coil and leucine zipper domains of C-terminal myosin binding subunit of myosin phosphatase for solution NMR studies

► We report cloning, expression, and purification of CC, LZ, and CCLZ domains of MBS. ► Applied methods result in high level protein expression of 15N and 13C labeled MBS polypeptides. ► SEC, CD, and 1H-15N HSQC demonstrate that expressed MBS CC domain is folded and exists as a dimer. ► Expressed MBS LZ domain is unfolded and non-stable at physiological pH. ► Expressed MBS LZ domain adopts folded and stable conformation at acidic pH 4.5.
