Preparation of Cholecystokinin Molecularly Imprinted Polymer Monolith and its Application in Solid Phase Microextraction and HPLC Analysis

The synthesis and preparation of molecularly imprinted polymer (MIP) monolith for selective recognition of cholecystokinin (CCK) were carried out by epitope imprinting technique. The MIP was synthesized by using CCK5 as template in a micropipette tip, and the polymerization conditions were optimized. The synthesized MIPs were characterized by scanning electron microscope (SEM) and infrared spectroscopy. Employing this SPME monolith as solid phase microextraction (SPME) column, an analytical method was developed for determination of CCK by coupling to high performance liquid chromatography with ultraviolet detection (HPLC/UV), and the parameters affecting SPME were optimized. The proposed method was successfully applied for the determination of CCK5 and CCK8 in spiked cerebrospinal fluid (CSF) simultaneously. The detection limit of CCK5 and CCK8 were 4.4 ng mL−1 and 16.0 ng mL−1, respectively, and the mean recoveries were 90.2% and 87.6% with the relative standard deviations of 3.2% and 3.9%, respectively. This CCK-MIP monolith column showed high specificity, excellent reusability and good applicability.

A MIP monolith for selective extraction of cholecystokinin (CCK) was prepared in a micropipette tip by epitope imprinting technique using CCK5 as template and methacrylic acid (MAA) as functional monomer.
