Anaplasma phagocytophilum in horses and ticks: A preliminary survey of Central Italy

Anaplasma phagocytophilum, the causative agent of granulocytic ehrlichiosis, affects several species of wild and domesticated mammals, including horses. In this work we compared direct and indirect methods to evaluate A. phagocytophilum presence in Central Italy: 135 sera were screened by IFA for A. phagocytophilum and other haemopathogens (Theileria equi and Babesia caballi). Each horse was also tested for A. phagocytophilum 16S rRNA with a nested-PCR technique. In order to examine the risk of A. phagocytophilum transmission, 114 ticks were examined for the presence of A. phagocytophilum by PCR targeting the 16S rRNA. The seroprevalence against A. phagocytophilum was 17.03% and 11 horses (8.14%) showed positive PCR results. The concordance rate of A. phagocytophilum detection between IFAT and PCR had a K value of 0.34.

RésuméAnaplasma phagocytophilum, l’agent causal de l’ehrlichiose granulocytaire, affecte plusieurs espèces de mammifères sauvages et domestiques, y compris les chevaux. Dans cette étude, nous avons comparé les méthodes directes et indirectes pour é evaluer la présence d’ A. phagocytophilum dans le centre de l’ Italie: 135 sérums ont été analysés par IFA pour A. phagocytophilum et pour autres pathogenès (Theileria equi et Babesia caballi). Tous les chevaux ont également été testé pour A. phagocytophilum 16S rRNA avec une technique de nested-PCR. Afin d’étudier le risque de trasmission d’ A. phagocytophilum, 114 tiques ont été examinées pour la présence de A. phagocytophilum par PCR pour le 16S rRNA. La séroprévalence de A. phagocytophilum a été de 17,03% et 11 chevaux (8,14%) ont montré des résultats positifs à la PCR. Le taux de concordance de détection de A. phagocytophilum entre IFAT et PCR avait une valeur K de 0,34.
