Streptomyces mobaraensis transglutaminase (MTG) is one of the most useful transglutaminases due to its rather broad substrate specificity and independence of Ca2+. To achieve efficient production of active-form MTG using Streptomyces lividans as a host, we created three vector constructs consisting of the signal peptide sequence (pld signal) derived from the phospholipase D gene of Streptomyces cinnamoneus, prepro-domain of S. cinnamoneus transglutaminase, and the sequence encoding mature MTG, and then generated three over-expressing S. lividans strains. We successfully demonstrated that S. lividans can be used as a host for the efficient production of mature, active-form MTG.
► Streptomyces mobaraensis transglutaminase is one of the most useful transglutaminases.
► The pld-signal and prepro-domain of S. cinnamoneus transglutaminase encouraged the secretion of MTG.
► We successfully produced a large amount of MTG using a genetically modified strain of S. lividans.
