► We developed and evaluated a fully automatable focus-reduction neutralisation test. ► Viral foci were visualized with immunoperoxidase-staining. ► Automated counting of foci was performed using ViruSpot software. ► The conventional Edmonston measles strain was used. ► The assay can be carried out with other types of viruses.
