Chinese hamster ovary cell lines selected for resistance to ebolavirus glycoprotein mediated infection are defective for NPC1 expression

► Selected CHO cell lines specifically resistant to ebolavirus GP-mediated infection. ► Host factors previously implicated in ebolavirus entry do not confer susceptibility. ► Resistant cells encode distinct mutations that knock out NPC1 protein expression. ► Expression of NPC1 restores infection. ► NPC1 mutations affecting cholesterol regulation but not protein expression were not selected.
