► An ECL peptide-based biosensor for prostate-specific antigen was fabricated. ► A helix peptide was employed as molecular recognition element. ► The detection limit for PSA was 0.038 ng/mL. ► The kinetic studies of peptide with PSA were performed. ► The method provides a promising strategy for detecting protein with simplicity.
