A reassessment of synchronous fluorescence in the separation of Trp and Tyr contributions in protein emission and in the determination of conformational changes

- An equation to predict synchronous spectra from excitation and emission data is derived.
- Synchronous spectra are unreliable to isolate Trp and Tyr contributions in protein spectra.
- For this purpose, a spectral deconvolution procedure gives superior results.
