Determination of phospholipase activity of PAF acetylhydrolase

► We describe a radiometric assay for determination of platelet-activating factor (PAF) acetylhydrolase activity. ► This convenient method is based on the release of [3H]acetate from [acetyl-3H]PAF. ► Separation between substrate and product is achieved using disposable cartridges. ► We also describe other approaches for measurement of PAF acetylhydrolase activity. ► Pros and cons of various methodologies are discussed.
