Optimization of a protease production process was studied in a bubble column bioreactor operated in fed-batch mode, using a newly isolated Pseudomonas sp. RAJR 044. The microorganism was cultivated in Czapek-Dox medium containing wheat bran and sucrose (as the sole carbon source). A mathematical model for the bioprocess was developed for optimization of production parameters where the root mean square error in modeling was found to be 2%. Genetic algorithm had been then used to perform the optimization task. The mathematical model was verified by experiment under optimized process conditions and the root mean square error in this case was found to be 3%. The genetic algorithm being an extremely flexible optimization tool had been used here for minimization of reaction time while maximizing product concentration.
