► Gly3157.42 and Ser3197.46 stabilize β2-AR in an inactive conformation. ► Occluded surface calculations show Gly3157.42 does not form a water binding site. ► Gly3157.42 acts as a “molecular notch” to stabilize Trp2866.48 in an inactive position. ► Ser3197.46 mediates hydrogen bonding interactions between Trp2866.48 and Asn3227.49.
