Use of human MonoMac6 cells for development of in vitro assay predictive of adjuvant safety in vivo

► We developed in vitro assay to predict in vivo toxicity of adjuvants. ► Pro-inflammatory cytokines were secreted by MM6 cells in presence of TLR agonists but not adjuvants. ► TLR agonists induced pro-inflammatory cytokines in vitro and febrile responses in rabbits. ► TLR agonists induced fever in rabbits was preceded by increased Prostaglandin E2 in plasma.
