► Synthesis of neutral lipid could be induced by cell-cycle arrest in conditional mutants of microalgae. ► Microalgae mutants blocked late in the cell cycle accumulate neutral lipids at the expense of polar lipid. ► Conditional microalgae mutants allow an efficient transition from biomass production to lipid accumulation.
