Galactans from the red seaweed Amansia multifida and their effects on inflammation, angiogenesis, coagulation and cell viability

Carrageenans are galactans known in the food industry as jellifying agents. Polysaccharides from Amansia multifida were previously extracted and precipitated at different volumes of acetone, obtaining three fractions (F1, F2 and FT). Chemical and spectroscopic analysis showed that the fractions were predominantly sulfated galactans. In anticoagulant activity tests FT showed greatest activity in the aPTT test with 200 μg (more than 240 s). Hemolytic activity showed that FT obtained the best lysis activity of the complement system, increasing by 42.3% at 50 μg. Of the three fractions analyzed, FT had the best output and anticoagulant and hemolytic effect, and was therefore chosen for in vivo inflammation assays. Inflammation tests with FT (50 mg/kg) to induce cell migration and IL-6 production showed an increase of 670.1% and 531.8%, respectively. Such data confirm its use as an inducer of inflammation in experimental animal models. FT exhibited antiangiogenic action with a score of 0.95 (100 μg/egg) and 1.45 (1000 μg/egg). The cytotoxicity assay showed cytotoxic action in all the fractions with 48 h exposure in a healthy 3T3 cell line and Hela tumor cell line, except fractions F1 and FT with 100 μg. This result leads us to questions their use as food, cosmetic and drug additives.
