A fraction of primitive “lymphoid” precursors retain plasticity for myeloid differentiation. In this issue of Immunity, Laiosa et al. describe that Notch-Delta signals can protect thymic precursors from reprogramming into the myeloid lineage, antagonizing the enforced myeloid transcription factors such as PU.1 and C/EBPα.
