Selective inhibition of the West Nile virus methyltransferase by nucleoside analogs

► We synthesized five unnatural nucleoside analogs. ► Two analogs can efficiently inhibit WNV methyltransferase (MTase) activity. ► The anti-WNV MTase compounds did not inhibit human mRNA MTase. ► The mechanism of selectivity is different from what was reported previously.
