Simultaneous detection and differentiation of four closely related sweet potato potyviruses by a multiplex one-step RT-PCR

► A multiplex RT-PCR was developed to detect four viruses infecting sweet potato. ► Four virus-specific forward and one common reverse primers were selected. ► The optimized assay reduced the cost of reagent and labor. ► The assay was validated using 300 samples to be reliable and sensitive.
