Quantitative Proton Nuclear Magnetic Resonance evaluation and total assignment of the capsular polysaccharide Neisseria meningitidis serogroup X

► This paper discusses a novel quantitative proton magnetic resonance method for capsular polysaccharide of X serogroup, a critical target for the next generation of meningococcal vaccines. ► The method has is linear, accurate, precise (under intermediate precision conditions) and rugged. ► The first total NMR assignment of the polysaccharide combining correlation experiments of 1H, 13C and 31P nuclei is reported. ► The quantitative NMR method and the NMR assignments discussed are valuable tools for further studies, especially in vaccine candidates.
