► NPC1L1 mediates cholesterol absorption in small intestine and liver. ► NPC1L1 recycles between PM and ERC in response to cholesterol levels. ► The N-terminal domain of NPC1L1 binds cholesterol not phytosterols. ► NPC1L1 and flotillins form cholesterol enriched membrane microdomains. ► Cholesterol is taken up by the endocytosis of these membrane microdomains.
