► Human infection with Necator americanus can be established reliably in a clinical setting. ► Experimental hookworm infection is safe and mostly well tolerated. ► N. americanus reduces the Th1/Th17 inflammatory celiac disease response to gluten. ► After hookworm infection, tolerance of gluten-derived antigens is enhanced. ► Celiac disease provides new insights into autoimmunity and how helminths regulate allergic and auto-inflammatory responses.
