► Mitochondrial uncoupling was induced by treatment with metabolic uncoupler, DNP. ► DNP significantly augmented CHOP contents and AMPK activity in 3T3-L1 adipocytes. ► DNP significantly decreased MCP-1 release in 3T3-L1 adipocytes. ► DNP reduced MCP-1 release via induction of ER stress and by activation of AMPK. ► JNK phosphorylation in 3T3-L1 adipocytes was decreased by treatment with DNP.
