► Most hematopoietic stem cells (HSCs) remain quiescent. ► Quiescence in HSCs is tightly regulated by many cell-cycle regulators. ► In most cases, ablation of such regulators in mouse HSCs results in loss of stemness. ► Future studies are required to translate the mouse knowledge to the clinical setting.
