► A novel process reduced the levels of proteins and polyphenols in tobacco. ► Levels of some tobacco smoke constituents were reduced, including aromatic amines. ► Aromatic amines might contribute to the bacterial mutagenicity of tobacco smoke particulate matter. ► The novel process reduced the bacterial mutagenicity of tobacco smoke particulate matter. ► The novel process did not affect in vitro mammalian genotoxicity of tobacco smoke particulate matter.
