► Common methods to prepare supported lipid bilayers are reviewed ► An overview of the force spectroscopy mode of AFM applied to bilayers is presented. ► The capacity of force spectroscopy to detect bilayer transitions is discussed. ► The effect of ionic strength and lipid chemistry on membrane mechanics is discussed. ► Emerging techniques and real-time AFM imaging is briefly introduced.
