► We evaluated molecular mechanisms of antipsychotics plus valproate combination. ► Antipsychotic plus valproate impact on Homer1a is different from individual effect. ► Our data confirm Homer1a region-specific modulation by antipsychotics. ► Homer1a expression is correlated with the degree of dopamine D2 receptors blockade. ► Homer1a is modulated in the cortex by dopamine and serotonin neurotransmission.
