► Two novel antimicrobial peptides (AMPs) active against Gram − and Gram + bacteria ► Arg instead of Lys residues improve AMP activity against Gram + bacteria. ► AMP mode of action against Gram + requires electrostatic binding but also deeply insertion in the membrane. ► Lys vs Arg residues relevant for AMP specificity vs broad spectra of action
