آشنایی با موضوع

آسیبDNA(به انگلیسی: DNA damage) بیشتر آسیب های DNA در اثر شرایط معمول زندگی ما روی می دهند مثلا جهش ها ممکن است در اثر تابش های طبیعی خورشید یا مصرف غذاهای کباب شده روی آتش یا استنشاق دود وآلاینده های هواایجاد شوند. جهش های اگزوژن ممکن است در اثر انواع عوامل ایجاد شوند. مثلا ممکن است در اثر مواد شیمیایی که به آنها جهش زا (موتاژن) می گویند باشد. چون دسته ای از این مواد شیمیایی سرطان زا هستند به آنها سرطان زا ( carcinogen)هم گفته می شود. گاز شیمیایی خردل که در جنگها استفاده می شوند نیز از عوامل شیمیایی جهش زای خطرناک است. برای نخستین‌بار محققان اهمیت آسیب به DNA را در تنظیم دقیق سیستم ایمنی ذاتی (Innate Immune System) انسان و در‌نتیجه توانایی تولید پاسخ التهابی ایده‌آل (Optimal) به عفونت‌ها و دیگر خطرات بیولوژیکی را نشان‌دادند و نتایج آن را درمجله‌ی Immunity به‌چاپ رساندند. این گروه تحقیقاتی در آزمایشگاه عفونت مولکولی پزشکی (MIMS) دانشگاه اومئوی سوئد، علاقه‌مند پی‌بردن به چگونگی تنظیم سیستم ایمنی ذاتی، اولین خط دفاعی بدن واینکه چگونه نقص در سیستم ایمنی بدن موجب بروز بیماری‌های عفونی و التهابی می‌شود، هستند. سیستم ایمنی بدن قبل‌از ایجاد پاسخ به‌صورت ساکن و در انتظار مورد حمله واقع‌شدن نمی‌باشد. حتی در غیاب عفونت، سیستم ایمنی بدن انسان دریک حالت ثابت هشدار (Constant State of Alert) قرار‌دارد. از میان واسطه‌های ایمنی بدن که به‌طور مداوم به میزان پایین تولید‌شده و سیستم ایمنی بدن را در‌حالت هشدار نگه‌می‌دارند، می‌توان از عواملی به‌نام اینترفرون‌های نوعI نام‌برد. وجود تعادل بسیار ظریفی درتولید این‌نوع اینترفرون برای سلامتی ضروری است؛ تولید ناکافی آن سبب زمینه‌ی ابتلا به عفونت‌های ویروسی می‌گردد در‌حالی‌که تولید بیش‌از‌حد معمول آن، منجر به بیماری‌های خود ایمنی و التهابی می‌شود. یکی‌از اهداف محققان پی بردن به فرآیندهای پیام‌رسانی کنترل‌کننده‌ی تولید اینترفرون نوع I و به‌طور خاص شناسایی «سیگنال‌های خطر» درون‌زاد (Endogenous) است که به‌طور مداوم باعث تولید پایه‌ای(Basal) اینترفرون می‌شوند و در‌نتیجه سیستم ایمنی بدن را در وضعیت «آماده برای حمله» نگه‌می‌دارند. سرنخ رسیدن به این هدف، بیماری نادر اما پیچیده‌ای به‌نام آتاکسیا تلانژکتازیا (AT) است. این بیماری با ویژگی‌های گوناگونی از‌جمله تخریب نورون‌ها، افزایش خطر ابتلا به سرطان، نازایی و شتاب‌ گرفتن روند سالمندی مشخص می‌شود. علاوه‌براین بیماران‌AT مستعد ابتلا به سندرم‌های مختلف خود ایمنی و التهابی هستند که در‌حال‌حاضر هیچ درمان قطعی و یا داروی خاصی برای این بیماری وجود ندارد. همزمان با این بررسی، با همکاری متخصصان بیمارستان Halmstad مشاهده‌شد که در سلول‌های بیماران‌AT میزان غیرطبیعی و بالایی از اینترفرون نوعI به‌صورت خود‌به‌خود و حتی در‌صورت عدم وجود عفونت تولید می‌شود؛ بنابراین چنین سلول‌هایی در مقایسه با افراد سالم که از عفونت جان سالم به در نمی‌برند، قادر به تولید پاسخ قوی‌تری بوده و از این رو قدرت محافظت بیشتری در برابر ویروس‌ها دارند. این مشاهدات، الهام بخش محققان MIMS جهت انجام ارزیابی‌های این ویژگی‌ در سطح مولکولی بود. با بررسی‌های انجام‌شده روی مدل ترمیم ژنتیکی‌شده‌ی (Genetically Engineered) موش، محققان قادر به کشف جزئیات بیشتری از عملکردهای پیام‌رسانی ایمنی شدند؛ و آنها برای اولین‌بار نشان‌دادند که شکست مولکول DNA) DNA Breaks Decipher) همان «پیام خطر درون‌زاد» است که باعث تولید پاسخ پایه‌ای اینترفرون نوع‌I شده و سیستم ایمنی بدن را در حالت هشدار نگه‌می‌دارد. مولکول‌DNA ما که حاوی تقریباً23000‌ژن است و تمام جنبه‌های فیزیولوژی ما را کنترل‌می‌کند، با ارزش‌ترین دارایی بدن ما است. مولکول‌DNA همیشه در‌معرض خطر مداوم آسیب ناشی‌از حـوادث نـرمال سلولی مانند همانندسازی (Replication (DNA، عوامل جهش‌‌زای متابولیک درون زاد یا عوامل مخربی مانند تشعشعات رادیواکتیو، نور فرابنفش و یا موادشیمیایی زیست‌محیطی قراردارد. علاوه‌بر‌این بسیاری از میکروب‌ها به DNA آسیب می‌رسانند؛ چه به‌طور مستقیم با قراردادن‌DNA خود درون DNA میزبان و چه با آزاد‌کردن مواد جهش‌زا(Mutagens) که ممکن‌است موجب آسیبDNA شوند. برای کاستن از این خطر مداوم، مقدار قابل‌توجهی از فعالیت‌های ترمیم و حفاظت سلول، به ایمنی و حفاظت‌DNA اختصاص داده شده‌است. در واقع قدیمی‌ترین و حفاظت‌شده ترین مولکول‌های پیام‌رسان در زندگی(حیات) یوکاریوتیک (Eukaryotic Life)، مولکول‌هایی هستند که به ترمیم شکست‌های‌DNA اختصاص یافته‌اند. با این حال در صورت ایجاد آسیب عمده در‌DNA، این نوع مولکول‌های پیام رسان باعث شروع مرگ برنامه‌ریزی شده‌ی سلولی می‌شوند؛ در نتیجه سلول اطمینان حاصل می‌کند که DNA آسیب دیده به سلول‌های دختر (Daughter Cell) انتقال نمی‌یابد.
