آشنایی با موضوع

هورمونهای اصلی بدن انسان را بر اساس ساختار مولکولی می‌توان به سه گروه تقسیم نمود: هورمون‌های استروییدی مانند هورمونهای جنسی، هورمون‌های آمینو اسیدی و هورمون‌های پپتیدی. گیرنده‌های هورمون‌های آمینو اسیدی در روی سلول‌های هدف قرار دارد و گیرنده‌های هورمون‌های استروییدی در درون سیتوپلاسم یا روی هسته قرار دارند. هورمونهای استروئیدی می توانند از غشای تمامی سلولها انتشار یابند ولی تنها در در سلولهای هدف با گیرنده های اختصاصی خود مواجه می شوند. سپس کمپلکس هورمون - گیرنده تحت تاثیر واکنش فعال شدن وابسته به حرارت و نمک قرار می گیرد. این واکنش منجر به بروز تغییراتی در اندازه و شکل و بار سطحی مولکول شده و آنرا قادر می سازد تا به کروماتین هسته متصل گردد. کمپلکس هورمون - گیرنده به یک ناحیه اختصاصی در DNA به نام عنصر پاسخ دهنده به هورمون متصل شده و ژنهای اختصاصی را فعال و یا غیر فعال می نماید. این هورمون ها دارای هسته ای هستند که از کلسترول مشتق می شوند. هورمون های قشر فوق کلیه (کورتیزول و آلدوسترون)، هورمون های تخمدان ها (استروژن و پروژسترون)، هورمون بیضه ها (تستوسترون) و هورمون جفت (استروژن و پروژسترون)- به طورکلی هورمون های غدد تولید مثل- و متابولیت های فعال ویتامین D جزو این دسته اند. تولید: این هورمونها اغلب در غده فوق کلیوی و غدد جنسی ساخته می‌شوند و کلسترول پیشساز طبیعی آنها است. در تولید اینها ابتدا کلسترول به پروژسترونها تبدیل می‌شود و سپس سایر هورمونها (آندروژنها، مینرالوکورتیکوئیدهاواستروژنها) تولید می‌شوند. کلسترول ۲۷ کربنه، پروژسترونها ۲۱ کربنه، گلوکوکورتیکوئیدهاو مینرالوکورتیکوئیدها ۲۱ کربنه، آندروژنها ۱۹ کربنه و استروژنها ۱۸ کربنه هستند. مکانیسم هورمون های استروئیدی: ‌وزن مولکولی هورمون های استروئیدی کوچک است و از طریق انتقال نفوذ یا حامل به سلول های هدف، هورمون ها را به سلول می رساند پس از اولین بار با گیرنده در سیتوپلاسم برای تشکیل یک گیرنده هورمون پیچیده، پیچیده در دمای مناسب و + مشارکت، تحول توانایی برای به دست آوردن از طریق غشای هسته ای. پس از اینکه هورمون وارد هسته، آن را به گیرنده در هسته متصل به تشکیل یک ترکیب. این مجتمع به سایت های خاص کروماتین متصل می شود، فرایند رونویسی DNA را در محل شروع یا مهار می کند، در نتیجه، ترویج یا مهار تشکیل mRNA، ایجاد یا کاهش سنتز پروتئین های خاص (به طور عمده آنزیم ها) برای دستیابی به اثرات بیولوژیکی آنها است. مولکول هورمون می تواند هزاران مولکول پروتئین را برای رسیدن به تابع تقویت هورمون تولید کند.
