فاکتور رشد عصب، (Nerve growth factor) یکی از فاکتورهای رشد با ساختاری پروتئینی است. فاکتور رشد مولکولهایی طبیعی با ساختار پروتئینی یا استروئیدی هستند. این فاکتورها در رشد و تمایز سلولهای زیادی دخیل هستند و نوعی سیتوکین هستند. سیتوکین‌ها دسته‌ای از مولکول‌های پروتئینی محلول در آب هستند که از یاخته های گوناگون و بیشتر در پاسخ به یک تحریک٬ ترشح می‌شوند و وظیفهٔ انتقال پیام بین یاخته ها را برعهده دارند. پی‌آمد حضور سیتوکین٬ تغییر در رفتار سلول‌های دارای گیرنده سیتوکین ترشح شده از جمله رشد٬ تغییر یا مرگ یاخته است. کنش و اثر سیتوکین تولید شده از یک سلول بیشتر بر سلول‌های پیرامون همان سلول را شامل می‌گردد ولی می‌تواند کنش‌سیستمی و اثر بر تمامی ارگانیزم را بدنبال داشته باشد. سیتوکین دارای اثر اتوکراین (تغییر در خود یاخته ترشح کننده) و پاراکراین (تغییر در دیگر یاخته ها) بوده و می‌تواند مانند یک هورمون اثراتی بر سلول‌های بسیار دورتر از خود نیز داشته باشد. نقش اصلی مثبت و منفی فاکتورهای رشد در تحریک تکثیر سلول‌ها و در تنظیم رشد سلولها در چندین سال اخیر به نحو فزاینده‌ای ارزیابی شده‌است. یکی از نخستین فاکتورهای رشدی که کشف شد، عامل رشد عصب در سال ۱۹۵۱ بود. این فاکتور رشد در رشد سلول‌های عصبی، بقا و فعالیت آنها مؤثر است. امروزه گروهی از عوامل رشد عصب تحت عنوان نوروتروفین شناخته شده‌اند که فاکتور رشد عصب اولین عضو این خانواده است. فاکتور رشد عصب یا NGF، فاکتور اختصاصی برای زنده ماندن نورون‌های حسی و سمپاتیک در طی تکامل سیستم عصبی می‌باشد. به دنبال قطع اکسون، سلول‌های شوان در قطعه انتهایی شروع به تولید NGF می‌کنند. NGF سبب تحریک مهاجرت سلول‌های شوان و کاهش دژنره شدن و افزایش ترمیم عملی پس از صدمه اعصاب محیطی و افـزایش رگ زائی می‌شود. . تجویز NGF سبب افزایش تعداد آکسون‌های میلین‌دار، ضخامت میلین و بلوغ بیشتر لایه‌های اندوتلیال می‌گردد. استفاده از پیوند اتوگرافت شریان و ورید رانی بعنوان کانال‌ راهنمای عصب به دلیل داشتنNGF در لایه‌های انتیـما و ادوانتیس ورید و شریان، موجب تسریع ترمیم اعصــاب محیطی می‌شود. . مقدار بالایNGF نه تنها ترمیم را تسریع می‌کند بلکه سبب توقف استحاله عصب نیز می‌گردد(7). IGF با تاثیر بر روی DNAسلول شوان، تولید فاکتور رشد عصب را افزایش می دهد. همچنین تحریک کننده قوی برای تزاید و تمایز سلول‌های شوان است. تجویز IGF-I در محل ضایعه عصب، سبب تقویت جوانه زدن آکسونی و افزایش تعداد آکسون‌ها در هنگام ترمیم عصب می‌گردد، و همچنین قطر رشته عصبی و ضخامت میلین را افزایش می‌دهد. مطالعات میکروسکوپ الکترونی نشان می دهد که IGF-I سبب افزایش تعداد میکروتوبول‌ها (که نشانه افزایش فعالیت در آکسوپلاسم است) می‌شود.
