آشنایی با موضوع

فلوئورسین ایسوتیوسیانات (به انگلیسی: FITC، fluorescein isothiocyanate) با فرمول شیمیایی (C۲۱H۱۱NO۵S) ترکیب شیمیایی با شناسه پاب‌کم ۱۸۷۳۰ است. که جرم مولی آن ۳۸۹٫۳۸۲ g/mol می‌باشد. برای انجام فلوسایتومتری لازم است که ابتدا سلول‌ها با فلوروکروم‌ها نشاندار شوند. از طرفی برای نشاندار کردناجزاء داخلی سلول باید اقدامات خاصی را انجام داد تا آنها در دسترس آنتی‌بادی یا ماده فلورسنت قرار گیرند. تعداد فلوروکروم‌های مورد استفاده برای فلوسایتومتری در طی سالیان متمادی مرتبا افزایش یافته است و این احتمال وجود دارد که در آینده، تعداد فلوروکروم‌های مورد استفاده برای آنالیز سلول‌ها بیش از این نیز افزایش یابدشناخت انواع فلوروکروم‌ها و آگاهی از مزیت‌ها و معایب هر کدام تنها راه استفادهبهینه از آنها برای دستیابی به مقاصد علمی تحقیقاتی است. امروزه پرمصرف‌ترین رنگ‌های فلوئورسان متصل به آنتی‌بادی‌ها در فلوسایتومتری بالینی، فلوئورسین ایزوتیوسیانات (FITC) و فیکواریترین (PE) می‌باشند. تمامی این رنگ‌های فلوئورسان در محدوده۴۸۸nm طیف‌های جذبی دارند. بنابراین، یک، تک طول موج تحریکی لیزری، می‌تواند تمامی این دو رنگ را تحریک کند. بر اساس پارامترهای تفرق نوری (FALS و SS) می‌توان gateهای الکترونیکی (محدوده‌ها) بدور دسته جات سلولی مورد نظر برای تجزیه با فلورسنت کشید، آنگاه فلوئورسانس سلول‌ها در نمونه می‌تواند بطور انتخابی تعیین شود و بصورت توزیع کثرت ترسیم گردد. کلیه اطلاعات توسط یک کامپیوتر که در یک سیستم گنجانیده شده است، جمع‌آوری، محاسبه و ذخیره می‌گردد. غالباً حجم مورد نیاز از نمونه موردآزمایش نیز خیلی کم و حدود ۱۰۰ میکرولیتر می‌باشد. بررسی فلوسایتومتریک هرچه زودتر باید انجام شود (پس از نمونه‌گیری)، بهرحال نمونه‌ها را می‌شود برای ۲۴ ساعت در دمای اتاق (مخصوص خون کامل) و یا ۴oC (مخصوص سلول‌های منونوکلئر جداشده) نگهداری نمود و سپس ایمونوفنوتایپینگ انجام داد. در هنگامی که نمونه‌ها برای بیشتر از ۲۴ ساعت نگهداری می‌شوند، اضافه کردن محیط کشت سلولی، مانند RPMI ضروری است. فلوروکروم ها برای انجام فلوسایتومتری لازم است که ابتدا سلول‌ها با فلوروکروم‌ها نشاندار شوند. از طرفی برای نشاندار کردن اجزاء داخلی سلول باید اقدامات خاصی را انجام داد تا آنها در دسترس آنتی‌بادی یا ماده فلورسنت قرار گیرند. تعداد فلوروکروم‌های مورد استفاده برای فلوسایتومتری در طی سالیان متمادی مرتبا افزایش یافته است و این احتمال وجود دارد که در آینده، تعداد فلوروکروم‌های مورد استفاده برای آنالیز سلول‌ها بیش از این نیز افزایش یابد شناخت انواع فلوروکروم‌ها و آگاهی از مزیت‌ها و معایب هر کدام تنها راه استفاده بهینه از آنها برای دستیابی به مقاصد علمی تحقیقاتی است. امروزه پرمصرف‌ترین رنگ‌های فلوئورسان متصل به آنتی‌بادی‌ها در فلوسایتومتری بالینی، فلوئورسین ایزوتیوسیانات (FITC) و فیکواریترین (PE) می‌باشند. تمامی این رنگ‌های فلوئورسان در محدوده۴۸۸nm طیف‌های جذبی دارند. بنابراین، یک، تک طول موج تحریکی لیزری، می‌تواند تمامی این دو رنگ را تحریک کند.
