آشنایی با موضوع

پلاسمون سطحی (SP) یک موج الکترومغناطیسی با قابلیت عملکرد بالا است که در فصل مشترک فلز – دی-الکتریک در سطح فلز در ناحیه طول موج نوری به دام می افتد. پلاسمون سطحی می تواند توسط موج فرودی تهییج شود که به این پدیده رزونانس پلاسمون سطحی (SPR) گویند. زمانی که این پدیده رخ می دهد، شدت نور بازتابیده به شدت کاهش می یابد. انهدام پلاسمون سطحی تهییج شده شامل تبدیل انرژی به فونون یا فوتون است. برای اینکه SPR رخ دهد باید Ksp=Kevan باشد. پژوهشگران از این روش استفاده کردند تا بتوانند اتم‌ها یا ذرات منفرد را شناسایی کنند. آنها یک چیدمان جدید ارائه کردند که در آن یک نانوذره پالادیوم را در منطقه فوکوس ایجاد شده توسط نانوآنتن، قرار دادند. برهمکنش میان طلا و نانوذره پالادیوم می‌تواند منجر به تولید LSPR شود به‌طوری که هر ذره‌ای که به نزدیکی این منطقه آورده شود عملکرد دی‌الکتریک ذره پالادیوم را تغییر می‌دهد. پرتو پراش یافته بوسیله این سیستم میکروسکوپ میدان تاریک ضبط شده و می‌توان با آن تغییر LSPR را رصد کرد. دستگاهوری SPR: چیدمان کرچمن در بیشتر دستگاههای SPR استفاده میشود و در شکل 3 ساختار آن مشاهده میشود. در این چیدمان، لایه ای از فلز (معمولا طلا یا نقره) در فصل مشترک دو محیط دی الکتریک قرار میگیرد. محیط 1 منشوری است که دارای ضریب شکست بالاتر است و محیط 2 با ضریب شکست پایین تر می تواند هوا یا محلول مورد نظر باشد. آنالیز با تکنیک SPR: زاویه SPR دارای رابطه خطی با ضریب شکست محلول در فصل مشترک است. زوایای SPR در محیط های با ضریب شکست متفاوت اندازه گیری شده اند. در شکل 6 ارتباط بین زاویه SPR و ضریب شکست نمایش داده شده است. اندازه گیری از طریق تکنیک SPR بدون نیاز به برچسب است، یعنی نیاز به مواد رنگی یا فلوئورسانس برای شناسایی سیگنال ندارد، برای مطالعه اجزای غذایی، مواد سمی یا توکسین ها، پاتوژن ها یا مواد بیماری زا، ویتامین ها، پروتئین های درمانی، نشانگرهای زیستی سرطان، آنتی بادی های تشخیصی، آلاینده های محیطی، شناساگرهای بیماری های قلبی، هورمون ها و الیگونوکلئوتید ها به کار می رود. سیستم های بر پایه رزونانس پلاسمون های سطحی بیش از دو دهه است که به صورت گسترده در کاربردهای مختلف به خصوص در حوزه علوم پزشکی مورد استفاده قرار گرفته است و این سیستم به صورت غیرتهاجمی و بر چسب آزاد به بررسی و شناسایی مولکول ها و اجزای مختلف می پردازد. در آینده توانایی انجام عملکردهای مختلف با استفاده از این سیستم نوری توسعه خواهد یافت.
