آشنایی با موضوع

ریزآرایه(به انگلیسی: Microarray) فناوری بررسی فعالیت ده ها، صدها و هزاران ژن و یا پروتئین در یک سطح کوچک ( یک لام میکروسکوپی و یا یک ریز ارایه در حد چند سانتی متر مربع) جهت مقایسه، مشابهت و یا بررسی تغییرات شامل، تغییر، کاهش، افزایش و یا عدم تغییر در ساختار و فعالیت ژن ها و یا پروتئین های نمونه با نمونه های شاهد. ریزآرایه هایDNA قطعات کوچک جامدی هستند که توالی های هزاران ژن متفاوت هرکدام روی مکان های خاصی از آن متصل شده یا ثابت شده اند. بخش پایه یا جامد اسلایدهای شیشه ای میکروسکوپی به اندازه دو انگشت کوچک دست انسان در کنار هم، هستند، البته بخش پایه می تواند به صورت تراشه های سیلیکون یا غشای نایلونی باشد. DNA روی این بخش پایه پرینت، نقطه گذاری و یا به صورت مستقیم ساخته می شود. نظم ژنها، یعنی این که بدانیم هر نقطه مربوط به کدام ژن است، بر روی تراشه بسیار اهمیت دارد، چرا که پژوهشگران بدین طریق به میزان بیان و. . . پی می برند. نقاط مذکور می توانند DNA، cDNA و یا الیگونوکلئوتید (به طول 5 تا 50 نوکلئوتید) باشند. حاصل بدست آمده ارزش بسیاری در تشخیص، تفکیک و ژنوتایپ عوامل عفونی؛ تغییرات مولکولی سلول ها و بافت ها و شناسایی عوامل مولکولی تغییر یافته در یک سلول، بافت و ارگان اسیب دیده با نمونه سالم و در نهایت استفاده گسترده از این فناوری در تحقیق، توسعه، پیشگیری، تشخیص و درمان را عملی می سازد. در واقع فناوری ریز ارایه نه تنها تحول بزرگی را در بهداشت و درمان ایجاد خواهد نمود بلکه در دهه اینده شاهد وارد شدن ان به بازار و کاربردهای گسترده در ازمایشگاه های تحقیقاتی و تشخیصی پزشکی خواهیم بود. انواع ریز آرایه ها عبارتند از: 1. ریزآرایه DNA: مثل ریزآرایه هایcDNA، ریزآرایه های اولیگونوکلئوتید و ریزآرایه هایSNP2. MMChipها: که در بررسی توده های میکروRNA کاربرد دارد. 3. ریزآرایه های پروتئین 4. ریزآرایه های بافتی 5. ریزآرایه های سلولی (به آنها ریزآرایه های ترانس فکشن نیز گفته می شود). 6. ریزآرایه های ترکیبات شیمیایی 7. ریزآرایه های پادتنی و 8. آرایه های کربوهیدراتی (گلیکوآرایه ها). سه نوع نمونه برای ساختن ریزآرایه هایDNA وجود دارد، دو نوع از آنها ژنومیک اند و نوع دیگر ترانسکریپتومیک است. در نوع اخیر سطوح mRNA اندازه گیری می شود. چیزی که این سه نوع را از یکدیگر متمایز می کند نوع DNAی ثابت استفاده شده برای ساختن ریزآرایه و نوع اطلاعات قابل استخراج از تراشه می باشد. DNAی هدف استفاده شده تعیین کننده نوع DNAی کنترل و نمونه مورد استفاده در محلول هیبریدیزیشن است. ریزآرایه ابزاری به صورت یک غشای کوچک یا اسلاید شیشه ای که دارای نمونه هایی از ژن های بسیاری که با طرح خاصی منظم شده اند، می باشد.
