توالی‌یابی | دانلود مقالات ISI توالی‌یابی | 1155 مقاله انگلیسی + ترجمه فارسی

آشنایی با موضوع

توالی‌یابی، در علم ژنتیک و بیوشیمی، به معنی تعیین ساختار داخلی یک بسپار (نظیر ترتیب نوکلئوتیدهای سازنده دی‌ان‌ای) است. نتیجه ی این فرایند، دستیابی به توالی ساختارهای مختلف اتمی در بسپار است که نمایانگر ساختار توالی مولکول در سطح اتمی میباشد. توالی یابی دی‌ان‌ای باید سه ویژگی داشته باشد ۱-مکمل بودن ۲-وارون بودن ۳-زوج نوکلئوتید بودن. انواع متنوعی از توالی یابی وجود دارد که عبارتند از: توالی‌یابی DNA توالی‌یابی RNA توالی‌یابی پروتئین توالی‌یابی پلی ساکارید دو روش برای تعیین توالی DNA ابداع گردید: 1) روش خاتمه زنجیره (Chain Termination squencing)2) روش تجزیه شیمیایی (Chemical Degradation squencing)اشکال عمده در تعیین توالی DNA دو رشته‌ای در مقایسه با DNAهای تک رشته‌ای در این است که حضور رشته مکمل در آزمایشات منجر به افزایش میزان خطاها در تعیین توالی می‌گردد. این خطاها به صورت مناطقی برروی ژل آشکار می‌شوند که در یک نقطه از ژل بیش از یک باند وجود دارد در نتیجه مشکل بتوان تصمیم گرفت که نوکلئوتید واقعی در این جایگاه کدام یک است. خطاهایی از این نوع تنها محدود به آنالیز DNA دو رشته‌ای نمی‌باشند و برخی از آنها یک مشکل عمومی در آزمایشات تعیین توالی می‌باشند. در صورت بروز این خطاها می توان برای رفع ابهام آزمایش را با یک پرایمر جدید با استفاده از رشته دیگر به عنوان الگو، تکرار کرد. در مورد توالی‌های بزرگتر از 700-600 نوکلئوتید حتی در صورت استفاده از DNA تک رشته‌ای در آنالیز توالی نیز همیشه نمی‌توان بدون بروز این خطاها محصولات واکنش‌ها را بر روی ژل الکتروفورز یا کاغذ کروماتوگرافی دقیقا از یکدیگر تفکیک نمود و البته این اندازه نوکلئوتیدی حداکثر طول توالی است که می‌توان توسط ترکیب ویژه‌ای از الیگو ـ پرایمر آنرا تشخیص داد. پیمایش با پرایمر ((Primer walking روشی دیگر برای تهیه الگو در آزمایشات تعیین توالی است. در این روش ابتدا یک پرایمر الیگونوکلئوتیدی که مکمل بخش انتهایی ناحیه شناخته شده یک توالی DNA است طراحی می‌گردد و پس با جفت کردن این پرایمر نشاندار با بخش مکمل آن، دنباله آن که در واقع بخش مکمل با ناحیه ناشناخته است سنتز می‌گردد و به این صورت کپی‌های تک رشته‌ای از ناحیه ناشناخته تهیه می‌شود که می‌توان در آزمایشات تعیین توالی از آنها استفاده کرد. البته می‌توان در همین روش به جای سنتز رشته جدید، با حذف آنزیمی توالی‌های موجود در فرودست ناحیه اتصال پرایمر به الگو مجموعه‌ای از قطعات ناشناخته تهیه کرد. یکی از روش هایی که در تاریخ علم تعیین توالی ژنوم کامل موجودات بسیار مهم شده است روش تعیین توالی شکاری ((Shotgun sequencing است که در اوایل کشف تعیین توالی از آن استفاده می‌شد در این روش DNAیی که قرار است توالی آن مشخص شود به طور تصادفی شکسته می‌شود و آنگاه قطعات حاصل در یک پلاسمید یا حامل M13 کلون می‌شوند. سپس از محل اتصال این قطعات به DNAی حامل همانند سازی در یک یا هر دو جهت آغاز می‌شود. فرایند تکثیر تا زمانی که مقدار DNAی تولید شده به 10ـ5 برابر DNA الگوی اولیه برسد ادامه می‌یابد و بعداً با استفاده از کامپیوتر هم پوشانی توالی‌های مختلف در این مجموعه DNA خوانده می‌شود و از این اطلاعات می‌توان برای تعیین توالی مولکول DNA اولیه استفاده کرد. هرچه تعداد این هم پوشانی‌ها که دارای نقاط شروع و پایان متفاوتی هستند و در جهتهای مختلفی نیز خوانده می‌شوند بیشتر باشد به همان میزان اطمینان از صحت توالی تعیین شده نیز بالاتر می‌رود. با تعیین توالی هر 2 رشته مولکول DNA به طور مجزا و با تائید مکمل بودن آنها می توان خطاهای تولید شده در زمان آزمایش یا در طی مرحله الکتروفورز را تشخیص داده و حذف کرد. امروزه روش های خودکار تعیین توالی تغییری اساسی در سرعت و جهت گیری پرو‍ژه‌های ژنومی ایجاد کرده است. از طریق این روش ها تا زمان نگارش کتاب توالی کامل ژنوم بسیاری از باکتریها چون مخمر Saccharomces cerevisiae و Caenorhabditis تعیین شده است و پروژه ژنوم انسانی نیز رو به کامل شدن است.
